I am getting a lot of trackback spam, and have set all trackbacks to auto-moderation. However, I would also like the wordlist to reject spams with certain words in the weblog address (to save me getting hundreds of emails a day).

It does not look like this is possible with the current spam protector plugin - if you set API comments to moderate, then the wordfilter is ignored.

Is it possible to do this, and if not, could this be considered for a future enhancement?

You are right, you can only set the API comments to "no special action" and then they would get rejected again.

In fact they do concur...hm, it's pretty hard to make the large spam protector filter all occasions and still not having an orverkill of option sets...
